چکیده

1: In recent years, capillary-based LightCycler® Systems have become an established standard for rapid and accurate real-time PCR. Based on innovative technology for heating/cooling and highly accurate analysis algorithms, they enable thermal cycling at high speed and with precisely controlled temperatures, characteristics directly related to the consistency and accuracy of scientific results. The recently developed multiwell (96/384) plate-based LightCycler® 480 Instrument goes one step further in providing enhanced PCR throughput and flexibility. Key to the system ́s speed and accuracy are a completely new designed thermal block and optical system which both minimize inter-well, inter-assay and inter-instrument variability. The LightCycler® 480 System uses a time-tested software algorithm for efficiency-corrected relative quantification and automated, meltingcurve based genotyping. Data on this poster will illustrate how the transfer from a capillaryto a multiwell-based LightCycler® System was technically achieved while maintaining speed and accuracy. (presentation of LightCycler® 480 System continued on P13 and P14)

MeDIP Real-Time qPCR has the Potential for Noninvasive Prenatal Screening of Fetal Trisomy 21

This study aimed to verify the reliability of the 7 tissue differentially methylated regions used in the methylated DNA immunoprecipitation (MeDIP) real-time quantitative polymerase chain reaction (real-time qPCR) based approach of fetal DNA in maternal blood to diagnosis of fetal trisomy 21. Forty pregnant women with high risk pregnancy who were referred after first or second trimester screeni...

Long-term surveillance of sulfate-reducing bacteria in highly saline industrial wastewater evaporation ponds

Abundance and seasonal dynamics of sulfate-reducing bacteria (SRB), in general, and of extreme halophilic SRB (belonging to Desulfocella halophila) in particular, were examined in highly saline industrial wastewater evaporation ponds over a forty one month period. Industrial wastewater was sampled and the presence of SRB was determined by quantitative real-time PCR (qPCR) with a set of primers ...

Measuring E. coli and bacteriophage DNA in cell sonicates to evaluate the CAL1 reaction as a synthetic biology standard for qPCR

We measured the impact of the presence of total Escherichia coli (E. coli) cellular material on the performance of the Linear Regression of Efficiency (LRE) method of absolute quantitative PCR (LRE qPCR), which features the putatively universal CAL1 calibration reaction, which we propose as a synthetic biology standard. We firstly used a qPCR reaction in which a sequence present in the lone gen...

Recent advances in quantitative PCR (qPCR) applications in food microbiology.

Molecular methods are being increasingly applied to detect, quantify and study microbial populations in food or during food processes. Among these methods, PCR-based techniques have been the subject of considerable focus and ISO guidelines have been established for the detection of food-borne pathogens. More particularly, real-time quantitative PCR (qPCR) is considered as a method of choice for...
